OBJECTIVE: To investigate the killing effect of hypocrellins B-photodynamic therapy (HB-PDT) to esophageal cancer cell line, and compareir with hematoporphyrin derivative (HpD), so as to disclose the superiorities and prospects of HB-PDT in the therapy of esophageal cancer. METHODS: Esophageal cancer cells were incubated in vitro with HB and HpD of different concentrations which dissolved in DMEM, and then irradiated by copper vapor laser with mixed wavelength light under saturated light dose. Cell survival rate of each concentration was measured by MTT assay after 24 hours' incubation. Accordingly, cell survival curves of different photosensitizer was described and the equation of each cell survival curve were analyzed, and finally, IC50-(50% inhibition concentration) of each photosensitizer was found. RESULTS: HB-PDT had the strong killing effect to esophageal cancer cell; the IC50 of HB was 34.16 ng/mL for esophageal cancer cell and the IC50 of HpD was 1526.03 ng/mL. There was a significant difference in the killing effect between the two photosensitizers, P=0.0001. CONCLUSIONS: HB-PDT has the strong photodynamic killing effect to esophageal cancer line; HB seems more effective photosensitizer than the first generation photosensitizer HpD.
