Associations of maternal HLA-G gene polymorphism, perinatal infections and smoking in the family with risk of congenital malformations in the fetus

ЦЕЛЬ ИССЛЕДОВАНИЯ

Изучить связь материнского полиморфизма гена HLA-G (rs41551813, rs12722477, rs41557518 и rs66554220), перинатальных инфекций и курения в семье с риском врожденных пороков развития у плода.

МАТЕРИАЛ И МЕТОДЫ

Обследованы 335 женщин, вынашивающих/родивших ребенка с врожденными пороками развития (ВПР), и 418 женщин без ВПР у плода/ребенка. Инфекционный статус беременных женщин оценивали, исходя из данных лабораторных тестов: микроскопического исследования (выявление бактериального вагиноза и вульвовагинального кандидоза, Treponema pallidum); иммуноферментного анализа и полимеразной цепной реакции (ПЦР) (выявление вируса простого герпеса I и II типа, цитомегаловируса и др.; Neisseria gonorrhoeae, Chlamydia trachomatis и др.). Фактор курение в семье оценивали как курение одного или обоих партнеров. Типирование полиморфных сайтов Thr31Ser (rs41551813, HLA-G*01:03) в экзоне 2, Leu110Ile (rs12722477, HLA-G*01:04) и 1597 delC (rs41557518, HLA-G*01:05N) в экзоне 3 гена HLA-G проводили методом асимметричной ПЦР. Полиморфизм гена HLA-G 14 bp Ins/Del (rs66554220) в 3’UTR области экзона 8 детектировали с помощью электрофоретического разделения продуктов амплификации в 7% полиакриламидном геле.

РЕЗУЛЬТАТЫ

Проведенное исследование показало, что перинатальные инфекции матери (OR=1,75 [1,31—2,34]) и курение в семье (OR=1,75 [1,31—2,34]) являлись основными факторами риска ВПР у плода (P<0,001). Материнская аллель 110Ile HLA-G (rs12722477, HLA-G*01:04) была дополнительным фактором риска ВПР у плода после введения поправки на перинатальные инфекции и курение в семье (OR=1,86 [1,13—3,05], P_c=0,03). Материнские полиморфные сайты rs41551813, rs41557518 и rs66554220 гена HLA-G значимо не влияли на риск ВПР у плода.

ЗАКЛЮЧЕНИЕ

Перинатальные инфекции матери, курение в семье и материнский полиморфизм гена HLA-G rs12722477 могут быть факторами риска врожденных пороков развития у плода. Наши результаты способствуют пониманию молекулярных механизмов формирования патологии в системе «мать — плод».

Association between cytokine gene polymorphisms and squamous cell lung cancer depending on the duration of smoking in men

Background. Squamous cell lung cancer (SCLC) is the most common form of lung cancer among men smokers. Mediators and products of inflammation can contribute to the initiation of carcinogenesis in smokers. The aim of study is to investigate the association between IL1B(rs1143634, rs16944), IL1RN (rs2234663), TNFA (rs1800629), IL6 (rs1800795), IL10 (rs1800896) genes and SCLC depending on the duration of smoking in men. Materials and methods. We studied 324 patients with SCLC and 168 healthy men smokers. The typing of the IL1RN(rs2234663) gene polymorphism has analyzed by polymerase chain reaction (PCR). Detection of the IL1B (rs1143634) and IL6 (rs1800795) polymorphisms have performed by restriction fragment length analysis, as a restriction enzyme TaqI was used. The IL1B (rs16944), TNFA(rs1800629) and IL10 (rs1800896) genes polymorphisms have determined through TaqMan-real-time PCR. Results. The study showed, the age, duration of smoking and the number of cigarettes smoked per day were the main risk factors for SCLC in men (p 0.0001). The IL6 -174G allele was detected an additional risk factor of SCLC at a smoking duration of less than 35 years (OR = 1.68; 95 % CI: 1.12-2.51; pcor = 0,04). No association of IL1B (rs1143634, rs16944), IL1RN (rs2234663), TNFA (rs1800629), and IL10 (rs1800896) genes with the risk of SCLC in men was identified. Conclusion. The duration of smoking and rs1800795 IL6 gene polymorphism may influence on the susceptibility to SCLC in men. Our results can be useful in understanding the molecular mechanisms of development of SCLC.

Исследовали содержание стероидных гормонов (эстрадиола, прогестерона) и специфических антител в сыворотке крови у 65 здоровых женщин и у 74 больных раком молочной железы в постменопаузе. Цель работы – выявить взаимосвязь между концентрацией стероидных гормонов и уровнем специфических антител в норме и при патологии. Впервые выявлены прямые корреляционные взаимосвязи между содержанием стероидных гормонов и соответствующих специфических антител. Эти взаимосвязи оказались наиболее выраженными у здоровых женщин. У больных раком молочной железы взаимосвязи между содержанием эстрадиола и уровнями антител к эстрадиолу сохранились, а между содержанием прогестерона и уровнями антител к прогестерону отсутствовали. Вероятно, на взаимодействие прогестерона и антител к прогестерону в сыворотке крови оказывают влияние соответствующие анти-идиотипические антитела, исследование которых еще предстоит выполнить. Таким образом, ранее высказанное предположение о влиянии антител к стероидным эндобиотикам, образующимся в естественных условиях, на биологические функции последних получило первое подтверждение.

Polymorphisms in DNA Repair and Xenobiotic Biotransformation Enzyme Genes and Lung Cancer Risk in Coal Mine Workers

Currently coal mining employs over 7 million miners globally. This occupational setting is associated with exposure to dust particles, heavy metals, polycyclic aromatic hydrocarbons and radioactive radon, significantly increasing the risk of lung cancer (LC). The susceptibility for LC is modified by genetic variations in xenobiotic detoxification and DNA repair capacity. The aim of this study was to investigate the association between GSTM1 (deletion), APEX1 (rs1130409), XPD (rs13181) and NBS1 (rs1805794) gene polymorphisms and LC risk in patients who worked in coal mines.The study included 639 residents of the coal region of Western Siberia (Kemerovo region, Russia): 395 underground miners and 244 healthy men who do not work in industrial enterprises. Genotyping was performed using real-time and allele-specific PCR.The results show that polymorphisms of APEX1 (recessive model: ORadj = 1.87; CI 95%: 1.01-3.48) and XPD (log additive model: ORadj = 2.25; CI 95%: 1.59-3.19) genes were associated with increased LC risk. GSTM1 large deletion l was linked with decreased risk of LC formation (ORadj = 0.59, CI 95%: 0.36-0.98). The multifactor dimensionality reduction method for 3-loci model of gene-gene interactions showed that the GSTM1 (large deletion)-APEX1 (rs1130409)-XPD (rs13181) model was related with a risk of LC development.The results of this study highlight an association between gene polymorphism combinations and LC risks in coal mine workers.

The retrospective epidemiological reseach of work conditions and level of oncological diseases rate in workers of Kemerovo «Koks» Inc. was conducted. At the same time the level weight of oncological diseases in the experiment group (88,9‰) reliably exceeds the control (51,0‰). The greatest difference were revealed in men (accordingly 90,3 and 33,2‰). The greatest relative risks were observed for the respiration organs cancer (4,3) and skin cancer (9,4) in men and for stomach cancer in women (7,0). It was found no relation between the indices of oncological diseases rate and the length of service in the control-cases from 3 to 25 years. The increase of service was more then 26 years. In contract, in experimental group with the length of service of 4-5 years the diseases rate of malignances is already twice times higher than in the control. With increasing of the length of service from 10 to 25 years, the sharp rise of diseases rate in experiment is observed (from 60 to 120‰). High indices of oncological deseases rate in workers with the length of service more than 26 years and not directly engaged in the production are possibly connected with not the industrial factors but with the changes caused by the age.

In the presented “case-control” study 670 residents of the Kemerovo Region subject to age, sex, ethnicity and smoking status were included. We formed two groups: 1) “Case” - 304 newly diagnosed lung adenocarcinoma patients undergoing a medical treatment in the Kemerovo Regional Oncology Center; 2) “Control” - 366 healthy donors of the Kemerovo Regional Center of Blood Transfusion. Statistical analysis were performed using SNPstats (http://bioinfo.iconcologia.net/SNPstats), «Statistica 10.0» (StatSoft, Inc., USA). A significant association obtained between the CYP1A1 (rs4646903Т>С), GSTТ1(del) and lung adenocarcinoma.

Estradiol is an estrogen derivative. It is capable to exert pathogenic influence on hormone dependent organs and tissues. Estradiol under certain conditions could stimulate the growth of benign and malignant tumors in the epithelium of the mammary gland, endometrium and cervix, epithelium and endothelium of mucous membranes (larynx, esophagus and rectum). The aim of this study was to obtain human single-chain idiotypic and anti-idiotypic antibodies against estradiol. A naive human phage library was screened. Seventeen (17) clones with a high-quality reaction to the estradiol-BSA conjugate have been selected as a result. Four singlechain antibodies had been decided on after evaluation of the amino acid sequences. The antibodies have been particular and had no analogues. The proteins have been expressed in E.Coli and isolated. The affinity of the antibodies in opposition to estradiol changed into measured through plasmon resonance. The most binding constant turned into determined for 119 antibodies.
