Quantitatively Relating Gene Expression to Light Intensity via the Serial Connection of Blue Light Sensor and CRISPRi

The ability to regulate endogenous gene expression is critical in biological research. Existing technologies, such as RNA interference, zinc-finger regulators, transcription-activator-like effectors, and CRISPR-mediated regulation, though proved to be competent in significantly altering expression levels, do not provide a quantitative adjustment of regulation effect. As a solution to this problem, we place CRISPR-mediated interference under the control of blue light: while dCas9 protein is constitutively expressed, guide RNA transcription is regulated by YF1-FixJ-PFixK2, a blue light responding system. With a computer-controlled luminous device, the quantitative relationship between target gene expression and light intensity has been determined. As the light intensifies, the expression level of target gene gradually ascends. This remarkable property enables sensor-CRISPRi to accurately interrogate cellular activities.

Significance Somatic mutations are the driving force of many age-related diseases such as cancer and hematopoietic failure. A challenge in the field is to evaluate health impact of somatic mutations prior to the appearance of disease symptoms. We describe a genetic tool named MASCOT (mosaic analysis system with Cre or Tomato) for mosaic analysis of somatic mutations that drive clonal hematopoiesis and lymphomagenesis. MASCOT can be applied to mosaic analysis of broad tissue types, and thus provides a valuable tool to aid functional dissection of somatic mutations in studies of development and disease.

The multiple antibacterial activities of the bladder epithelium

Abstract: The urinary tract is subject to frequent challenges from the gut microflora. Indeed, up to 40% of women will experience at least one urinary tract infection (UTI) during their lifetime. Uropathogenic Escherichia coli (UPEC) contribute to an overwhelming majority of these cases and they typically initiate UTIs by invading the superficial epithelium that lines the bladder lumen. In addition to serving as an effective barrier to noxious agents found in urine, bladder epithelial cells (BECs) play a key physiological role in regulating bladder volume to accommodate urine flow. UPEC appear to coopt this latter property to circumvent this normally impregnable epithelial barrier. However, in spite of this shortcoming, recent studies suggest that BECs possess several immune mechanisms to combat bacterial invasion including expulsion of invading bacteria back into the bladder lumen following infection. These antibacterial activities of BECs are triggered and coordinated by sensory molecules located on the epithelial cell membrane and within the cells. Although, they are the primary targets of microbial attack, BECs appear to be equipped with a diverse repertoire of defense schemes to fend off many of these microbial challenges.

LAT-mediated TCR signaling is required for optimal function and clonal expansion but not long-term survival and homeostatic turnover of DETCs (LYM7P.610)

Abstract Dendritic Epithelial T Cells (DETCs) are generated exclusively in the fetal thymus and maintained in the skin epithelium throughout life. DETCs have limited antigenic specificity due to their exclusive usage of a canonical TCR. While the requirement of TCR in DETC development has been clearly demonstrated, the role of TCR in DETC function and lifelong maintenance is still poorly defined. To investigate the role of TCR signaling in DETCs, we used a γδ lineage specific and inducible Cre allele to delete the gene encoding Linker Activation in T cells (LAT), an essential signaling molecule downstream of the TCR. We found that LAT-mediated TCR signals in DETCs were required for proper activation of effector genes and optimal clonal expansion. The function of DETCs in wound healing was also dependent on LAT. Lack of proper TCR signaling prevented DETCs from efficient production of growth factors and cytokines involved in wound healing. Surprisingly, we found that long-term maintenance of DETCs in the skin was not affected by deletion of LAT. These findings demonstrate that LAT-mediated TCR signaling is required for optimal function and clonal expansion of DETCs and implies a LAT-independent mechanism for long-term survival and homeostatic turnover of DETCs in the skin epithelium.
